It was developed by a group at the University of Texas Medical Branch as a membrane-permeable, selective NNMT inhibitor. It is not a SARM and not a peptide.

Interest in the enzyme came first. A 2014 Nature paper found that NNMT expression was raised in the white fat and liver of obese and diabetic mice, and that lowering NNMT in those tissues with an antisense oligonucleotide protected mice against diet-induced obesity, with higher energy expenditure and higher fat-tissue levels of S-adenosylmethionine and NAD+ (Kraus et al., 2014). That work used a genetic tool, not a drug, but it made NNMT a target, and small-molecule inhibitors such as 5-Amino-1MQ followed.

In a 2018 study, diet-induced obese mice given an NNMT inhibitor from this series showed lower body weight and white fat mass and lower plasma cholesterol than vehicle-treated animals over the treatment period (Neelakantan et al., 2018). A 2019 study in aged mice reported that the inhibitor increased muscle stem cell activity and larger muscle fibres after an experimental injury (Neelakantan et al., 2019). Both studies used subcutaneous injection in mice over a few weeks, the same laboratory produced both, and neither was designed to assess long-term toxicity. A 2021 study from a collaborating team developed an assay for the compound in rat plasma and urine and measured its behaviour in rats after intravenous and oral administration (Awosemo et al., 2021). There are no human trials, no human pharmacokinetic data and no safety data in people, and the compound has never been approved as a medicine. It is frequently sold alongside claims that nothing published supports. Nothing above is a claim about this product.

In chemical terms, PubChem (CID 950107) records 5-amino-1-methylquinolinium under CAS number 685079-15-6, with the formula C10H11N2+ and a mass of 159.21 for the ion. The plus sign matters: the ring nitrogen carries a permanent positive charge, so the substance cannot exist on its own as a neutral powder and is always supplied as a salt paired with a negative counter-ion. Its systematic name is 1-methylquinolin-1-ium-5-amine.

This listing

Manufacturer: Thoroughbred Labs

Compound: 5-Amino-1MQ

Form: Capsules

Pack size: 60 capsules

Strength: the manufacturer does not state the amount per capsule on the label, so we do not list one

Batch certificate of analysis: shown where Thoroughbred Labs supplies one for the batch we hold

5-Amino-1MQ in the UK: buying, legal status and what to check

Searches such as "5-Amino-1MQ UK" and "5 amino 1mq UK buy" ask whether a UK shop may sell it and how to judge a listing. 5-Amino-1MQ is not a licensed medicine in the UK, has not been assessed by the MHRA, and is not an authorised food supplement. The published record for each route is thin and should be read carefully.

The two mouse efficacy papers gave the compound by subcutaneous injection, so they say nothing direct about capsules. The only published oral data come from the 2021 rat study, which found the compound in plasma after oral administration and reported an oral bioavailability of about 38 per cent in rats, meaning a sizeable fraction of the orally administered amount reached the circulation in those animals (Awosemo et al., 2021). Rat figures cannot be converted into human figures, and no study has measured oral absorption in people.

Because 5-Amino-1MQ is a small molecule rather than a chain of amino acids, it is not broken down by stomach and gut proteases in the way oral peptides are, which is why the "peptide" label sometimes attached to it in search results is misleading. Its permanent charge is a different consideration, since charged molecules often cross gut membranes less readily than neutral ones.

Otherwise the formats differ in measurement. A capsule holds a weighed blend of active powder and filler, so consistency depends on even mixing; a solution depends on the compound staying dissolved and intact. Full thymosin beta-4 weighs about 4,963 g/mol, three and a half times as much. The full-length thymosin beta-4 protein went further, into clinical programmes for wounds, the cornea and cardiac repair, including a placebo-controlled intravenous tolerability study in healthy volunteers. Those studies used the full protein by injection or local application, not a capsule, and none produced an approved medicine. No trial has compared the two peptides, so neither can be ranked above the other. It was developed by GlaxoSmithKline and Ligand as a candidate for lipid disorders. It is not a SARM, although it is often sold alongside them, and it is not a peptide either: it is a small synthetic molecule built around a thiazole ring.

PubChem lists it as CID 9803963, CAS number 317318-70-0, molecular formula C21H18F3NO3S2, molecular weight 453.5 and InChIKey YDBLKRPLXZNVNB-UHFFFAOYSA-N. The systematic name, 2-[2-methyl-4-[[4-methyl-2-[4-(trifluoromethyl)phenyl]-1,3-thiazol-5-yl]methylsulfanyl]phenoxy]acetic acid, contains a sulphur atom in a thioether link; that sulphur is where the body oxidises the molecule, which matters for drug testing (see below).

What the published research reports

In a 2008 study in mice, GW-501516 given alongside treadmill training increased running endurance compared with training alone, which led the authors to describe the compound as an exercise mimetic; without training it produced no endurance change in that study (Narkar et al., 2008). A 2012 trial in 268 people with low HDL cholesterol reported that, compared with baseline, HDL rose and LDL cholesterol and triglycerides fell over twelve weeks (Olson et al., 2012); the trial was short and not designed to assess long-term safety. Development was abandoned in 2007 after two-year carcinogenicity studies in rats and mice found tumours in multiple organs. Those toxicology results were presented at a conference and are not indexed on PubMed, so they are not cited here, but they are the most important fact about this compound. GW-501516 activates PPAR-delta; SR9009 was described in a 2012 paper from the Scripps Research Institute as a synthetic agonist of REV-ERB-alpha and REV-ERB-beta, nuclear receptors that help drive the daily rhythm of clock genes (Solt et al., 2012). In that study, SR9009 was given to mice by intraperitoneal injection; the mice showed altered circadian activity and changes in metabolic gene expression, and diet-induced obese mice lost more fat mass than controls. Because it is orally available and mimics ghrelin at the pituitary, it stimulates the release of the body's own growth hormone rather than supplying the hormone directly. PubChem records the free base under CAS number 159634-47-6, with formula C27H36N4O5S, molecular weight 528.7 and InChIKey UMUPQWIGCOZEOY-JOCHJYFZSA-N.

Merck studied MK-677 in the 1990s and 2000s. Chapman and colleagues gave it by mouth once a day to 32 healthy volunteers aged 64 to 81, over study periods of two and four weeks. Growth hormone pulses grew taller without becoming more frequent, and serum IGF-1 climbed into the range seen in young adults; fasting glucose also rose, prolactin went up by 23 per cent while staying within the normal range, and cortisol did not change (Chapman et al., 1996).

The longest trial randomised 65 healthy adults aged 60 to 81 to MK-677 or placebo for up to two years, with its main end points assessed at one year (Nass et al., 2008). Fat-free mass rose by an average of 1.1 kg on MK-677 and fell by 0.5 kg on placebo, yet strength and function did not improve. On the other side of the ledger, body weight rose by 2.7 kg on MK-677 and 0.8 kg on placebo, fasting glucose climbed by an average of 0.3 mmol/L, insulin sensitivity fell and cortisol rose. Increased appetite, which eased after a few months, and transient mild leg swelling and muscle pain were the commonest side effects. MK-677 has never been licensed as a medicine. The molecule was first described in a 2011 paper from Radius Health, which described it as a potent, orally bioavailable, non-steroidal SARM and reported its characterisation in several preclinical models of anabolic androgen action (Miller et al., 2011). Those models were animal experiments, not human studies.

PubChem records it under CAS number 1182367-47-0, formula C20H16ClN5O2, molecular weight 393.8 and InChIKey XMBUPPIEVAFYHO-KPZWWZAWSA-N. The systematic name, 2-chloro-4-[[(1R,2S)-1-[5-(4-cyanophenyl)-1,3,4-oxadiazol-2-yl]-2-hydroxypropyl]amino]-3-methylbenzonitrile, shows two defined stereocentres and no steroid ring system, and vosilasarm is listed among its synonyms.

The only published human trial is a Phase 1 dose-finding study in patients with hormone-receptor-positive metastatic breast cancer, reported in 2022 (LoRusso et al., 2022). Raised AST, ALT and bilirubin were among the most frequent adverse events, grade 3 or 4 adverse events occurred in 16 of the 22 patients, and the investigators identified a maximum tolerated dose. Two blood markers moved as expected for an androgen: sex hormone-binding globulin fell in every one of the 18 women tested and prostate-specific antigen rose in 16 of 20, which the investigators took as evidence that the receptor was engaged. The trial assessed tolerability and tumour response, not muscle or strength. Outside trials, case reports describe cholestatic liver injury in people who had taken products sold as RAD-140 (Leung et al., 2022), and hypogonadism in a man whose supplements contained RAD-140 together with other undeclared substances, so no single compound could be blamed (Chong et al., 2024). The legal answer is narrow. RAD-140 has no MHRA marketing authorisation, so it is not a licensed medicine in the UK; it is not an authorised food supplement; and it therefore cannot lawfully be sold for human consumption. Free Muscle sells it to adults aged 18 or over for laboratory research only. WADA prohibits it at all times under section S1 (anabolic agents), and buyers are responsible for the rules that apply to them, including any anti-doping code. Of 44 products bought online as SARMs in 2016 and analysed with WADA-approved procedures, only 23 (52 per cent) contained a SARM, 4 contained no active compound, 11 contained substances not on the label, and the measured amount matched the label in just 18 (41 per cent) (Van Wagoner et al., 2017). The published research does not map neatly onto any of them. BPC-157 studies in rats mostly gave the peptide by injection, with some Zagreb experiments adding it to drinking water. Thymosin beta-4 work used intravenous infusion, eye drops, gels applied to skin and injection in animals. Nobody has published a measurement of either peptide in human blood after a capsule was swallowed, so the search "BPC 157 capsules vs injection" has no answer in human data. The routes used in each study are set out in BPC-157 capsules vs injection: what the studies used.

The obstacle for a swallowed peptide is chemical. Stomach acid unfolds proteins and pepsin begins cutting them, then pancreatic enzymes and the peptidases lining the small intestine finish the job, and an intact peptide would still have to cross the gut wall. The Zagreb group reports that BPC-157 withstands human gastric juice in its own tests; nothing comparable has been reported for the TB-500 fragment, whose two adjacent lysine residues are typical cleavage points for trypsin.

As materials, dry powder in a sealed shell or vial generally keeps better than peptide in water, and a capsule's content depends on how evenly the peptide was blended with filler. TB-500 is built on residues 17 to 23 of thymosin beta-4, the stretch of the parent protein that binds actin, with an acetyl group capping its start. At under 1,000 g/mol the fragment is smaller than BPC-157 at 1419.5 g/mol.

Evidence base

The two literatures grew in different ways. BPC-157 research is concentrated in one university network and is almost entirely rodent and cell work, with no randomised human trial published. Thymosin beta-4 has been studied by many independent laboratories and taken into sponsored clinical programmes using the full protein, none of which has yet led to an approved medicine. No study has ever compared BPC-157 and TB-500 directly, so neither can be ranked above the other.

Regulation and sport

In the UK both are unlicensed research compounds without MHRA, EMA or FDA approval. Their WADA sections differ: BPC-157 is in S0 because it has no approval for human therapeutic use, while TB-500 falls in S2 because the list names thymosin beta-4 and its derivatives. The practical result is identical, as both are prohibited at all times. Strict liability applies: an athlete answers for whatever a sample contains.

Laboratories have published detection methods for each peptide. For TB-500, the Ghent doping control laboratory identified Ac-LKKTETQ by high-resolution mass spectrometry, synthesised it as a reference material and suggested a plasma and urine detection route. For BPC-157, the Salt Lake City laboratory validated a urine method with a limit of detection of 0.1 ng/mL, showed the peptide remained stable in urine for at least four days, and added a stable metabolite as a second marker to improve specificity. Neither study followed people after exposure, so no human detection window has been established for either peptide. SLU-PP-332 switches on the ERR family, which governs how many mitochondria cells build. SR9009 has since shown what a longer record can reveal. In 2019 an independent group bred mice lacking both REV-ERB proteins and found that SR9009 still reduced cell viability and changed metabolism and gene transcription in their liver cells and stem cells, so its effects cannot all be credited to REV-ERB (Dierickx et al., 2019). Despite the name, these orphan nuclear receptors have nothing to do with the oestrogen receptor; in muscle and heart they control how many mitochondria cells build and how much fuel they oxidise. It came from the laboratories of Thomas Burris and John Walker, working at Saint Louis University and partner institutions, and its first full characterisation was published in 2023. In 2024 the group followed with diet-induced obese and ob/ob mice, reporting higher energy expenditure and fatty acid oxidation, less fat mass and improved insulin sensitivity. One author declared a co-founder role in a company developing such compounds. In a 2023 study in the American Journal of Pathology, 21-month-old mice treated for eight weeks showed reversal of age-related albuminuria, podocyte loss and inflammatory markers in the kidney. The doping control laboratory at Ghent University examined a TB-500 formulation by high-resolution mass spectrometry and identified the N-terminally acetylated 17-23 fragment of thymosin beta-4 in it. In the same year the racing laboratory of the Hong Kong Jockey Club described TB-500 as a veterinary preparation whose key ingredient was that acetylated seven-residue peptide, and published a method for finding it and its metabolites in horse urine and plasma. TB-500 meets neither framework, so the only basis for this listing is supply for laboratory research to adults aged 18 and over, not for human consumption. No thymosin beta-4 medicine holds a UK marketing authorisation. Thymosin beta-4 and its derivatives, with TB-500 named as an example, are listed under section S2 (peptide hormones, growth factors and related substances) of the WADA Prohibited List and are prohibited at all times in sport. Anyone subject to anti-doping rules should consult the current list. The published work on thymosin beta-4 used routes that bypass digestion: intravenous infusion in the healthy-volunteer study, eye drops and topical gels in the corneal and skin programmes, and injection in most animal models. No study has measured whether any intact TB-500 peptide reaches the blood of a person after a capsule is swallowed.

The reason is digestive chemistry. The stomach's pepsin and the pancreatic and brush-border peptidases of the small intestine exist to break peptide bonds, and a short linear chain such as LKKTETQ, rich in lysine residues that trypsin cleaves next to, is an ordinary substrate. Crossing the gut wall would be a second, separate hurdle.

As materials, the formats differ in two practical ways. Stability: dry powder, whether in a capsule or a vial, keeps better than peptide dissolved in water, where hydrolysis and microbial growth begin. Uniformity: a capsule's content depends on how evenly a small amount of peptide was blended with filler before filling, while a solution's depends on how accurately it was made up and measured out. None of this implies that any format produces an effect.
