Despite the name they do not bind oestrogen. They regulate the expression of genes for mitochondrial biogenesis, fatty acid oxidation and oxidative metabolism, and their activity rises in skeletal muscle in response to endurance exercise. SLU-PP-332 was developed at Saint Louis University, which is where the SLU prefix comes from, as a pan-agonist that switches on all three receptors. It was designed as a tool compound to test whether pharmacological activation of ERRs could reproduce part of the adaptive response to aerobic training.\u003c\/p\u003e\n\u003cp\u003eThe first pharmacology paper appeared in 2023, so the entire literature is only a few years old and comes largely from the discovering group and its collaborators. Follow-up work has explored the compound in mouse models of obesity and metabolic syndrome, heart failure and kidney ageing, and a successor compound, SLU-PP-915, has since been described. SLU-PP-332 has never entered a clinical trial. There is no human pharmacokinetic, safety or efficacy data of any kind, and it is not approved as a medicine anywhere in the world. Authors of both papers declared shareholdings in a company developing ERR-based therapeutics.\u003c\/p\u003e\n\u003cp\u003eIn a 2026 paper the same group described a chemically distinct successor, SLU-PP-915, and stated plainly that SLU-PP-332 improves aerobic performance in mice but lacks oral bioavailability; the newer molecule was characterised because it is active by mouth. In that paper SLU-PP-332 was given by intraperitoneal injection, and it too carries a declared conflict of interest: the senior author is named as inventor on the university's intellectual property covering SLU-PP-915 and holds shares in companies developing ERR agonists. There is no human data of any kind, no human pharmacokinetic study, and no safety study in people. It is one of the newest compounds in this market and one of the least characterised. The developers themselves describe SLU-PP-332 as lacking oral bioavailability, which is why they went on to make SLU-PP-915. No study has measured what swallowed SLU-PP-332 does in an animal or a person, so we make no comparison of forms on effect.\u003c\/p\u003e\n\u003cp\u003eThe oral-peptide argument does not apply here. SLU-PP-332 is not broken down by digestive proteases, because it is not a peptide; the developers report a bioavailability problem without, in the abstract, giving its cause. SLU-PP-332 switches on transcription factors, the ERR receptors, that drive genes for mitochondrial energy production. 5-Amino-1MQ (5-amino-1-methylquinolinium) works on an enzyme instead: it inhibits nicotinamide N-methyltransferase (NNMT), which methylates nicotinamide and in doing so draws on the cell's pools of NAD+ and the methyl donor SAM.\u003c\/p\u003e\n\u003cp\u003eThe key 5-Amino-1MQ paper came from the University of Texas Medical Branch.
