TB-500 (Thymosin Beta-4)

TB-500, also sold as thymosin beta-4 or Tβ4 and sometimes written TB500, is a synthetic peptide based on the actin-binding region of thymosin beta-4, a 43-amino-acid protein present in almost every cell of the body. It is studied in animal and cell models of wound repair and cell migration, and no version of it is an approved medicine. This page lists every brand of TB-500 stocked at Free Muscle, compared on pack size, form and batch testing, with links to the research and to the certificate for each batch.

What is TB-500?

Thymosin beta-4 is the body's main actin-sequestering protein. By binding G-actin it regulates how cells build their internal skeleton, which in turn affects cell migration, blood vessel formation and the inflammatory response. The product sold as TB-500 is not the full protein. Anti-doping chemists who characterised commercial TB-500 found it to be a short, N-terminally acetylated fragment covering residues 17 to 23 of thymosin beta-4, the segment that carries the actin-binding motif (Esposito 2012). That distinction matters, because most of the published biology concerns the full 43-residue protein rather than the fragment.

Full-length thymosin beta-4, international non-proprietary name timbetasin, has been given to people in a few small trials. An intravenous study in 40 healthy volunteers, with placebo in each cohort, found adverse events infrequent and mild or moderate, none serious, after single and then repeated administration over 14 days (Ruff 2010). A phase 2 trial of eye drops in nine people with severe dry eye found less discomfort and corneal staining than with vehicle drops, on 12 treated eyes against six (Sosne 2015). Nothing has been approved.

The fragment sold as TB-500 has no published human data at all, and the US FDA has stated that it identified no human exposure data for it. It is not a licensed medicine in the UK, the EU or the United States. Thymosin beta-4 and its derivatives, with TB-500 named as an example, are prohibited at all times under WADA class S2 (peptide hormones, growth factors and related substances).

PubChem records TB-500 under CID 62707662 and CAS number 885340-08-9: the N-acetylated seven-residue fragment Ac-LKKTETQ, formula C38H68N10O14, molecular weight 889.0 g/mol. Full-length thymosin beta-4 is a separate entry, CID 16132341 and CAS 77591-33-4, with the formula C212H350N56O78S and a molecular weight of about 4,963 g/mol.

What the published research reports

A 1999 study reported that topical or intraperitoneal thymosin beta-4 accelerated closure of full-thickness skin wounds in rats, with increased collagen deposition and keratinocyte migration (Malinda 1999). A 2004 study in Nature reported that thymosin beta-4 activated integrin-linked kinase, promoted cardiac cell migration and survival, and improved cardiac function after experimentally induced myocardial infarction in mice (Bock-Marquette 2004). A 2005 review summarised the protein's actin-sequestering role and its wound-repair activity across animal models (Goldstein 2005). All of this used the full protein, in animals, at doses and routes with no relation to any capsule product. The human studies of the full protein, an intravenous safety study and a small eye-drop trial (Ruff 2010; Sosne 2015), did not test the TB-500 fragment or any oral form, and no thymosin beta-4 product has been approved.

The other body of literature is analytical. The Ghent anti-doping laboratory identified the acetylated fragment in a TB-500 product, synthesised it as a reference and set out an LC-MS approach to detecting it in urine and plasma (Esposito 2012). After a single administration to horses, a Hong Kong racing laboratory found the fragment and its metabolites in both urine and plasma (Ho 2012).

A French racing laboratory noted that products sold online claim to contain either the fragment or the full protein, and showed that a synthesis impurity could be detected in a horse's plasma after one administration of a thymosin beta-4 product (Delcourt 2025). It is a peptide, not a steroid and not a SARM: it has no steroid ring structure and does not act on the androgen receptor.

Identifier

Thymosin beta-4 (parent protein)

Length

43 amino acids

Molecular formula

C212H350N56O78S

Molecular weight

about 4,963 g/mol

PubChem CID

45382195

InChIKey

UGPMCIBIHRSCBV-UHFFFAOYSA-N

Actin-binding segment

residues 17 to 23, LKKTETQ

What the published research reports

A 2012 review by Goldstein and colleagues summarised the peptide's properties and the clinical programmes then under way in wounds, the cornea and cardiac repair. A 2010 randomised, placebo-controlled study gave intravenous pharmaceutical-grade thymosin beta-4 to healthy volunteers as single and repeated doses and reported no serious adverse events across the range tested; it was a tolerability study, not an efficacy study, and it used injection rather than a capsule. The doping control laboratory at Ghent University examined a TB-500 formulation by high-resolution mass spectrometry and identified the N-terminally acetylated 17-23 fragment of thymosin beta-4 in it. In the same year the racing laboratory of the Hong Kong Jockey Club described TB-500 as a veterinary preparation whose key ingredient was that acetylated seven-residue peptide, and published a method for finding it and its metabolites in horse urine and plasma. It is not a controlled drug either. UK law turns on how a product is presented: anything offered to treat or prevent a condition is regulated as a medicine, and anything offered to be eaten must meet food law. TB-500 meets neither framework, so the only basis for this listing is supply for laboratory research to adults aged 18 and over, not for human consumption. No thymosin beta-4 medicine holds a UK marketing authorisation. Thymosin beta-4 and its derivatives, with TB-500 named as an example, are listed under section S2 (peptide hormones, growth factors and related substances) of the WADA Prohibited List and are prohibited at all times in sport. The published work on thymosin beta-4 used routes that bypass digestion: intravenous infusion in the healthy-volunteer study, eye drops and topical gels in the corneal and skin programmes, and injection in most animal models. No study has measured whether any intact TB-500 peptide reaches the blood of a person after a capsule is swallowed.

The reason is digestive chemistry. The stomach's pepsin and the pancreatic and brush-border peptidases of the small intestine exist to break peptide bonds, and a short linear chain such as LKKTETQ, rich in lysine residues that trypsin cleaves next to, is an ordinary substrate. Crossing the gut wall would be a second, separate hurdle.

As materials, the formats differ in two practical ways. Stability: dry powder, whether in a capsule or a vial, keeps better than peptide dissolved in water, where hydrolysis and microbial growth begin. Uniformity: a capsule's content depends on how evenly a small amount of peptide was blended with filler before filling, while a solution's depends on how accurately it was made up and measured out. None of this implies that any format produces an effect. A 2010 randomised, placebo-controlled study gave intravenous pharmaceutical-grade thymosin beta-4 to healthy volunteers as single and repeated doses. Adverse events were infrequent and mild or moderate, and none was serious. It was a safety study, not an efficacy study, and it used injection rather than a capsule. In 2012 the Ghent doping control laboratory analysed a TB-500 product by high-resolution mass spectrometry and identified the acetylated 17-23 fragment of thymosin beta-4, Ac-LKKTETQ, rather than the whole protein. The same year the Hong Kong Jockey Club's racing laboratory described TB-500 as a veterinary preparation built around that acetylated fragment and published a method that found it, and its breakdown products, in urine and plasma from horses given a single administration. The anti-doping work on the Ac-LKKTETQ fragment used horses given the veterinary preparation, not an oral form. No published study has measured whether either peptide reaches the blood intact after being swallowed. The general obstacle is digestion: stomach and intestinal proteases cleave peptide chains, and a 43-residue protein offers them many sites. Even the 7-residue fragment is a target, and the Hong Kong group's method looked for its metabolites as well as the parent peptide. For Yakka Strength products we show a certificate of analysis only where the manufacturer supplies one for the specific batch we hold. A generic PDF with no batch number is not a batch certificate, and we do not present one as such. Email us with the product name and we will send what we hold, or say plainly that there is none. That section applies in and out of competition, across every sport that has adopted the World Anti-Doping Code, including testing by UK Anti-Doping. Under strict liability an athlete is responsible for any prohibited substance in their sample, whatever the label said.

Detection methods have been published for more than a decade. The Ghent laboratory proposed an LC-MS/MS strategy for the Ac-LKKTETQ fragment in plasma and urine in 2012, and the Hong Kong racing laboratory reported confirmation of the fragment at 0.01 ng/mL in equine urine and 0.02 ng/mL in equine plasma, together with its metabolites, after a single administration to horses. Those limits come from horse samples. No published study has set out how long TB-500 remains detectable in human urine, so the accurate statement is that it is detectable and the window is not established.

Storage, stability and handling

No stability study of TB-500 capsules has been published, so the expiry or best-before date printed by the manufacturer, if present, is the limit to work to. General peptide chemistry still identifies the risks. Moisture allows peptide bonds to hydrolyse and makes powder clump and capsule shells soften. Heat accelerates every degradation reaction, and light adds to it. Full thymosin beta-4 also contains a methionine residue, which oxidises readily, whereas the Ac-LKKTETQ fragment has none.

Keep the tub closed somewhere cool and dry, away from direct light, radiators and damp rooms. If it is refrigerated, let it reach room temperature before opening so condensation does not form inside, and leave any desiccant sachet in the tub. Write the opening date next to the batch number. Handle the capsules as a laboratory reagent: gloves for opened capsules or loose powder, well away from food and drink, and out of reach of children and animals. Dispose of waste as chemical waste, not in household bins or drains. The side facts panel reads TB500, 250 mcg per capsule, sixty capsules per container, "Made in the UK", so 15 mg per pot as labelled; the back panel carries the Yakka Strength Ltd distributor line. The brand's product page calls the contents a synthetic peptide and no more: it does not say whether the pot holds the acetylated 17-23 fragment that laboratories have found in products sold as TB-500 or full-length thymosin beta-4, so the labelled figure cannot yet be tied to a specific molecule.

Yakka Strength Ltd (company 13284431, registered since 2021 in County Durham) is chiefly a gym equipment and sports nutrition retailer.
